The focus of this paper is to summarize various structures of proteins, various funcions of protein molecule in the cell, the link between molecular structure of proteins and their function, and how proteins as a whole contribute to health and/or disease. Studies conducted in this field have shown that there are four categories of protein structure. These are the primary structure, secondary structure, tertiary structure, and quaternary structu re. The amino acids are made up of a backbone section which is available in the different types of amino acids, together with a side chain for each residue (Nelson, L. & Cox, M. 2008). Since the carbon atom remains bound to the four categories, one isomer would happen in the biological protein. In this case, the molecules would be chiral. Glycine fails to be chiral due to the fact that it has a side chain that is an atom of hydrogen. The amino acids are bonded by a covalent bond chain which are referred to as the peptide bonds (Brown EC et al. 2004). Whenever the chain is short, such a chain is called a peptide. The chains that are long are referred to as proteins or polypeptides. In many cases, the peptide bonding is established in between the one amino acid carboxyl group together with the amino group. The bonding of peptides happens in the reactions of condensation, which involve the loss of a molecule of water. Primary structure of proteins. The primary structure proteins is the amino acid polypeptide chain sequence. The basic structure is combined together using the covalent bonding just like the case of the peptide bonds, which are often established in the translation process of proteins. The two ends of a polypeptides are referred to as the C-terminus. In this, the carboxyl terminus, and the N-terminus are the amino terminus (Mahn K et al. 2005). In order to count the residues, one would begin at the N-terminus, which involves the terminus where the group of amino acid fails to be involved in the peptide boding (Nelson, L. & Cox, M. 2008). This type of structure is identified by the corresponding gene to the protein molecule. A particular nucleotide sequence in DNA is normally shifted to the mRNA that is interpreted by ribosome in a process referred to as translation (Mahn K et al. 2005). In this case, the protein sequence is specific to the particular protein, and identifies the structure and the roles of the protein molecule. Protein sequence could be determined through tandem spectrometry tandem mass, and Ednan degradation (Nelson, L. & Cox, M. 2008). In most cases, it is always interpreted directly from the gene sequence through the utility of the genetic code. The post-translational modifications including the formation of disulfide, glycosylation, is considered as a section of the primary structure hence cannot be identified from the specific gene. For instance Insulin has fifty one amino acids classified in two chains, one having thirty one amino acids, and the other one having twenty amino acids (Mahn K et al. 2005). Receptors in Intestinal Muscles The experiment was conducted in order to gain better understanding of the function of the receptors in the guinea pig ileum. For this reason, various agonists and antagonists were used and the muscle reaction was monitored. The results of our experiment are summarized in the following table. As we can see acetylcholine and hexamethonium both have a triethylamine at one end and a straight chain of carbons. The basic difference is that hexamethonium has two tertiary amines, one on each end of the chain, whereas acetylcholine has the group -O-C(=O)-CH3 on one end. According to the SAR theory (Structure Activity Relationship) similar molecules in structure tend to have similar biological activity. As we know, both acetylcholine and hexamethonium bind to the nicotinic receptor, the first one to trigger a response and the second one to prevent acetylcholine from binding. Hexamethonium, having two active groups, can probably bind more easily to the receptor, effectively blocking the acetylcholine action. b) Histamine and mepyramine have less similarities in structure. Both of them have three nitrogen and an aromatic ring. Histamine has the two nitrogen inside the aromatic ring whereas mepyramine has only one nitrogen bound in the ring. Both compounds bind to the H1-Histamine receptor, to trigger different reactions. The difference in structure can be explained by the different action of the two compounds. Histamine causes contraction of the muscle and mepyramine causes its relaxation. The drugs tested were classified as agonists and antagonists. Acetylcholine: Acts as neurotransmitter. It binds on the muscarinic and nicotinic receptors and causes muscle contraction. Histamine: Is also a neurotransmitter. It binds on the H1-Histamine receptor and causes smooth muscle contraction. Nicotine: It acts on the nicotinic cholinergic receptors and mimics the neural transmission. It stimulates the muscle, then blocks stimulation. Isoprenaline: Although isoprenaline was apparently an antagonist, it is actually a selective agonist for the ÃŽÂ ²- adrenergic receptors that causes muscle relaxation. It is a sympathomimetic drug that mimics the effect of stimulating the postganglionic adrenergic sympathetic nerves. Hexamethonium: It is a nicotinic antagonist and a ganglionic blocker. It binds to the nicotinic cholinergic receptors and blocks the actions of acetylcholine or cholinergic agonists. It has no effects on muscarinic (mAch) receptors. Mepyramine: It is a histamine H1 antagonist and targets the H1- Receptor. Although it was believed to be an antagonist merely to block the actions of endogenous histamine without activating the receptors, it has recently been classified as an inverse agonist decreasing the spontaneous activity of gp-H1r. It also inhibits histamine induced inositol phosphate (InsP) production and intracellular calcium mobilization. It causes a marked decrease in the maximal response to histamine at high concentrations. Atropine: It is a competitive antagonist for the muscarinic cholinergic receptor (mAch). It binds to the receptor without activating it, thus blocking the actions of endogenous acetylcholine or exogenous agonists. a) The drug in this experiment were acting on three receptors. H1-Histamine receptors, muscarinic (mAch) receptors and nicotinic (nAch) receptors. Each agonist was acting on a different receptor and that is apparent from our results. When using an antagonist that blocked a specific receptor it only inhibited the action of the drug acting on that particular receptor, and had no effect on the rest of the drugs. b) The receptors were obviously located on the surface of the muscle, so that the access of the drugs would be possible. The first apparent antagonist which turned out to be an agonist was isoprenaline. It acts on the ÃŽÂ ²- adrenergic receptors causing muscle relaxation and antagonized all the three agonists who acted on different receptors. This type of antagonism is called a physiological antagonist and describes the interaction of two drugs who cause opposing actions in the body and tend to cancel each other. In this case, the isoprenaline acts on the ÃŽÂ ²- adrenergic receptors and causes relaxation of the muscle, whereas the agonist act on the histaminic, nicotinic and muscarinic receptors and cause contraction of the muscle. The second apparent antagonist was mepyramine, which acts on the histamine receptor and blocks the action of histamine. It has recently been classified as an inverse agonist, causing muscle relaxation. This type of agonists show selectivity to the resting state of the receptor. Atropine acts on the muscarinic receptors and blocks their action. Thus it prevents acetylcholine from binding to the receptor and stimulating it. Nicotine though activates the nicotinic receptor that apparently has nothing to do with atropine. The reversal of nicotine action indicates the presence of inhibitory postganglionic (terminal) neurones, which respond to stimulation of their ganglion-cells by inducing relaxation of the gut. It is also suggested by other experiments [Phillis York, 1968] that an intermediate type of receptor is involved. Assuming specificity of the antagonist these studies are explained by a non-classical cholinergic receptor with mixed pharmacological properties. Such receptors are the newest members of the nicotinic acetylcholine receptor (nAChR) family, encoded by the ÃŽÂ ±9/ÃŽÂ ±10 subunits, that possess a combined nicotinic-muscarinic sensitivity. Barium Chloride is a water soluble salt. Once in contact with the muscle it induces release of intracellular stores of calcium, and causes the contraction of the muscle. If barium chloride comes in contact with sodium sulphate it loses its potency. That is explained by the chemical reaction between the two compounds. Molecular equation: BaCl2(aq) + Na2SO4(aq)> BaSO4(s) + 2NaCl(aq) ionic equation: Ba+2(aq) + 2Cl-(aq) + 2Na+(aq) + SO4-2(aq) > BaSO4(s) + 2Na+(aq)+2Cl-(aq) These reactions show that once in contact with sodium sulphate, the barium chloride dissolves into BaSO4 which is an insoluble substance and NaCl. Thus, it can no longer act on the muscle. That type of antagonism is called Chemical Antagonism and it refers to the situation when two substances combine in solution; as a result, the effect of the active drug (in this case the barium chloride) is lost. The drugs were tested on guinea pig ileum which is a smooth muscle.

Computer Science    I have always been interested and intrigued by computers, ever since using a BBC when I was very young. Since then I have become fluent in writing BASIC and more recently I have learnt HTML, the language of the World Wide Web on which I have my own wesite. I use computers for most of my exam coursework such as Solving Equations Using Numerical Methods for Pure Maths 2 and also for recreation.    My A-level studies are Mathematics, Further Mathematics, Physics and Design Technology. I have always been fascinated by numbers and this interest is continuing, hence my choices in A-level subjects. I enjoy the application of Mathematics to Physics. In Design Technology I can be creative and at the same time use my knowledge of Mathematics and Physics to help solve problems.    I am applying for the course of Computer Science because I would like to increase my knowledge of this subject. I hope that in the future I will be able to create applications for computers that will help people to save time and money.    For a career, I would like to be involved in the fast developing computer industry and more particularly with the Internet. I think it has immense potential and I would like to be able to develop the Internet to make it help the lives of day to day people. In this way I would also like to help make computers more like humans with for example voice operated user interfaces.    For my University course I am seeking sponsorship from several companies associated with computers and computing.    I am an active member of my Sixth Form being a prefect with supervisory responsibilities. I hold the positions of both sixth form committee member and House Captain for my school house. I have performed in the VI form pantomime, The Wizard Of Oz, for the local primary school children. When my school took part in BBC Newsround?s national mock election I was the Party Leader for the Liberal Democrat Party. I designed posters on my computer and helped mastermind the campaign in the school. This involved speaking in a four-way debate between the three party leaders and an audience of interested sixth formers and staff. My hard work paid off and I won my seat for the Sixth Form with an overwhelming majority.

Taboo

What is taboo? Taboo is human activity that is forbidden based on moral judgment. Anything that is not accepted by society or by your peers is considered taboo. This perfectly awful word has so many meanings ,yet no matter what race, religion, or culture your in it means forbidden or banned. No matter how hard you try not to be taboo you are. So is being taboo part of human nature, or is it how we try to stand out from everybody else and show individuality? Are you taboo, do you know someone who is taboo?There is no taboo that is universal, but some occur in the majority of societies. The dietary practices of some people are considered taboo, for instance some are vegetarians, kosher diets, cannibalism, or meat-eaters. Judaism prohibits the eating of bats, frogs, crocodiles, or snakes. Cow or beef is taboo to be eaten by vegetarian, Hindus, or Zoroastrians. Dog meat is taboo in the United States, they are considered part of the family. While in China dogs are bred for their meat, one example of this is the Chow Chow.Prince Philip of the United Kingdom once said â€Å"If it has four legs and is not a chair, has wings and is not an aero plane, or swims and is not a submarine, the Cantonese will eat it. † Once again everybody is taboo. I personally love to eat frog and beef. It is taboo in my eyes not to eat meat ,but that is because I am a meat-eater. The way people show or deal with their body function, such as burping, flatulence, defecating, and urinating are all forms of taboo. In the United States it is considered taboo to defecate or urinate in public.While in India, they teach to drink cow urine and eat cow feces. We all try to avoid doing things we think are taboo. People deal with things they think are different in different ways. Some turn their heads when they are faced with something or someone they find ugly or repulsive. The way people stare with added expressions when they see something different then what they consider normal, anything from a personal choice to birth defects. Even in relationships there is and will always be taboos. Sex outside of arriage, adultery, miscegenation, and incest are all forms of taboos in relationships. In the United States adultery and incest are very taboo, but in Wodaabe ,men of Niger steal each others wives even if they already have one. So adultery is taboo here over there it is accepted and condoned. Miscegenation was taboo in the United States until 1967 when they said it was unconstitutional. Taboo is in the politics of everyday living. For the United States fascism, communism, and anarchism are all taboo.While in China and Vietnam both accept and have a communism government. In early German the Nazism was taboo because of the fascism. Taboo was brought to the English language by Captain James Cook, and English explorer who visited Tonga. A famous American author Henry Miller said â€Å" whenever a taboo is broken, something good happens, something vitalizing. Taboos after all ar e only hangovers, the product of diseased minds, you might say, of fearsome people who hadn’t the courage to live and who under the guise of morality and religion have imposed these things upon us. So do not shun or avoid , accept everybody’s taboos because when they look at you they see all your taboos to them. Taboos are everywhere and in everything, because if there wasn’t then this would be a boring world to live. Could you imagine if everybody was doing the same thing, everything going the same direction ,no disorder, or no rebellion it would be very bland. In the end no one is perfect, and even if it might be considered taboo, whatever it is. The taboos we have or don’t have make us all unique and special.

Tragedy and the Common Man in Hamlet

Katelyn Stoll Professor Hall English 102 11 November 2009 â€Å"Tragedy and the Common Man† in Hamlet Arthur Miller notes that, â€Å"The tragic feeling is evoked in us when we are in the presence of a character who is ready to lay down his life, if need be, to secure one thing—his sense of personal dignity† (1). This characteristic seen in most tragedies is definitely evident in the character of Prince Hamlet in Shakespeare’s play Hamlet. The moment that Hamlet learns from the ghost that Claudius has committed regicide, his goal becomes clear: he has to avenge the death of his father by murdering his uncle. Hamlet could not stand idly by while the assassin of his saintly father had an affair with his mother Gertrude and lied to the people of Denmark. However, Hamlet’s tragic flaw prevents him from taking action quickly. During the course of the play, the prince notes that he has yet to perform any action against his uncle Claudius, and he wonders why this is. The character of Hamlet is prone to reasoning and long soliloquies, not action; this, in my opinion, is his tragic flaw. The apparition of the late Hamlet informs his son that Claudius, the current king of Denmark, poisoned him. Upon hearing the news, Hamlet is enraged and swears to take revenge against his usurping uncle. Almost immediately he is ready to lay down his life to correct what has been done, and he now has a â€Å"†¦willingness to throw all he has into the contest, the battle to secure his rightful place in his world† (3). It is at this moment in the play that Hamlet takes on the role of the familiar tragic hero and acts accordingly. He was displaced from the life that he knew and loved and was not awarded with his rightful position in society. Hamlet should be the king of Denmark if what the ghost told him is true; not only is Hamlet not the king of Denmark, but also his mental health is constantly being called into question. He is losing ranks in society awfully quickly, and part of Arthur Miller’s definition of the tragic hero is that the hero strives to evaluate himself justly. His tragic flaw does not allow him to regain his personal dignity, however, and Hamlet becomes frustrated over time because of this. He either takes too much time thinking everything through, or he reacts impulsively and violently when the situation does not call for it. This is seen when Hamlet accidentally stabs Polonius to death, thinking him to be a spy. His tragic flaw is not knowing when or how to act aggressively, and it really costs him in the end. According to Miller, â€Å"For, if it is true to say that in essence the tragic hero is intent upon claiming his whole due as a personality, and if this struggle must be total and without reservation, then it automatically demonstrates the indestructible will of man to achieve his humanity† (4). He argues that the tragic play has a lot more to offer the spectator than just a sad or unfortunate ending. Shakespeare’s play, Hamlet concludes with the deaths of Gertrude, Laertes, Hamlet and Claudius. The point of this play, however, is not that four people died, but that Hamlet was finally able to avenge the death of his father. Although this was not a perfect victory for Hamlet, he was able to attain his goals, and this demonstrates the will of man (even the common man) to secure his sense of personal dignity. The thrust for freedom is the quality in tragedy which exalts† (3). The conclusion of Hamlet is both a wonderful and depressing one. In one sense, Hamlet is not a tragic hero, because he was able to overcome his tragic flaw and slay Claudius. In another more realistic sense, however, he perfectly fits the description of the tragic hero because he does not live long enough to see the benefits of his actions. Hamlet is never able to evaluate himself justly, and that was his main objective. In the tragic view the need of man to wholly realize himself is the only fixed star, and whatever it is that hedges his nature and lowers it is ripe for attack and examination† (3). Hamlet perfectly adheres to the definition of the tragic hero of Arthur Miller, because of his need to regain his personal dignity, his tragic flaw preventing him for achieving this, and a tragic ending in which his goals are never realized.
